Mosley’s most notable film role was as the lead character in Gordon Parks’ biography of the musician Leadbelly, which he starred in in 1976.
 Mosley stated in an article in the November 1982 issue of Ebony magazine that this was his favorite role.
From 1980 through 1988, Mosley starred on Magnum, P.I. He played Tom Selleck’s friend, helicopter pilot Theodore “T.C.” Calvin, who runs his own tourist tour company, Island Hoppers. In season five of Las Vegas, he played the millionaire friend of Montecito owner A.J. Cooper (Tom Selleck)
